xPONENT® Basic Training Course

 

This one day course covers instrument routine use as well as maintenance and troubleshooting exercises like understanding calibration and verification reports. Software instruction will include building protocols and batches, run batches and perform data analysis using the xPONENT® software package.

Protein Development Classroom Trainings

 

Module 1

 

This is a two-day course and covers skills for developing application specific immunoassays on the xMAP® platform. The practical session consists of coupling capture antibodies to paramagnetic MagPlex® Microspheres (two step carbodiimide coupling method), determining bead recovery, running a coupling confirmation assay, and performing a cross-reactivity test, including capture sandwich immunoassay format. An in-house assay will be used for the practical part of the course. The theoretical session will cover topics like coupling antibodies to Luminex microspheres, identifying components of antibody structure, developing capture sandwich immunoassay assay format, technical tips, assay development best guidelines, and advice on buffers and consumables. The course includes troubleshooting information on coupling and assay development.

Module 2

 

Module 2 is a two-day course and covers skills for developing application specific immunoassays on the xMAP platform. The practical session consists of coupling antigen to paramagnetic MagPlex Microspheres (two step carbodiimide coupling method), determining bead recovery, performing a coupling confirmation assay, and running an indirect immunoassay format. An in-house assay will be used for the practical part of the course. The theoretical session will cover topics like coupling different molecules (i.e. antigens, peptides, small molecules), developing different assay formats (i.e. indirect detection or serology assay, competitive inhibition immunoassay), identifying different coupling modification methods, developing an indirect immunoassay assay format, technical tips, assay development best guidelines, and advice on buffers and consumables. The course includes troubleshooting information on coupling and assay development.

Nucleic Acid Development Classroom Trainings

 

This two-day course covers skills for developing application specific nucleic acid assays on the xMAP platform. The practical session consists of coupling amino-modified oligonucleotides to paramagnetic MagPlex Microspheres, determining bead recovery, running a coupling confirmation assay, and running a multiplex direct DNA hybridization assay. An in-house assay will be used for the practical part of the course. The theoretical session will provide detailed information on developing MagPlex-TAG™; based assays. Different assays formats will be covered i.e., Allele and Target Specific Primer Extension (ASPE/TSPE), Oligo Ligation Assay (OLA), and Ligation Dependent Amplification (LDA). The attendees will gain knowledge of which application is best suited for their specific need and how to troubleshoot assays. They will receive tips on probe and primer design and advice on buffers and consumables.
