Upon administration of a pneumococcal vaccine, antibody response can achieve measurable concentrations in 2 to 5 weeks.  The efficacy of the vaccination is generally assessed by measuring pre- and post-vaccination antibody titers.  A 2-fold increase, or greater, is expected in most adults¹.  Published literature considers that immunoglobulin G (IgG) response varies by serotype and can be impacted by age and health status¹ ².  There is currently no documented agreement on specific post-vaccination antibody concentrations needed to establish protection from pneumococcal infection.

Assay Description

The Luminex Pneumococcal Immunity Panel utilizes Luminex xMAP® multiplexing technology to provide a quantitative assessment of IgG following administration of a pneumococcal vaccine.

xMAP® Pneumo14 contains the following pneumococcal serotypes:

Laboratory developed enzyme-linked immunosorbent assays (ELISA) are used by major reference laboratories and hospital laboratories to measure antibody response to vaccination.  Each ELISA measures a single serotype on one plate requiring a total of 14 plates to test 14 serotypes.  By contrast the xMAP® Pneumo14 assay allows for the simultaneous measurement of all 14 serotypes from a single patient sample in a single well.  This significantly reduces the amount of labor and time to results versus traditional methods.

Inter-Laboratory Reproducibility

Since the assay is usually executed with pre- and post-vaccination samples, reproducibility is of utmost importance.  This assay demonstrates superior reproducibility in the most stringent inter-laboratory scenarios.  In this example, 80 blinded samples run at two different laboratories showed an average correlation coefficients of 0.96.
