FlexmiR Product Overview

The FlexmiR™ microRNA products combine the power of xMAP® technology with Exiqon’s Locked Nucleic Acids™ (LNAs) to achieve best in class specificity using a quick, simple workflow.  The FlexmiR MicroRNA Human Panel, FlexmiR MicroRNA Mouse/Rat Extension Panel, and FlexmiR Select, in conjunction with the FlexmiR MicroRNA Labeling Kit and FlexmiR MicroRNA Control Set, provide methods that qualitatively and quantitatively measure the expression of the miRNA sequences listed in the miRBase Sequence database using total RNA samples without need for RNA size fractionation or amplification. 

Locked Nucleic Acids
LNA™, a technology owned by Exiqon A/S, is a conformationally restricted nucleic acid analogue, in which the ribose ring is “locked” with a methylene bridge connecting the 2’-O atom with the 4’-C atom. Incorporation of LNA monomers into an oligonucleotide capture probe greatly increases the affinity of the oligonucleotide for its complementary RNA target leading to a significant increase in stability and specificity of the duplex.

Currently many researchers use planar arrays for broad panel screens and qRT-PCR or Northern Blot for small, focused screens. FlexmiR Select allows for quantification of up to 50 miRNA targets simultaneously. Together, FlexmiR and FlexmiR Select offer researchers the ability to understand miRNA expression on a single platform.

Simple yet powerful.
The fast, simple workflow of FlexmiR Select uses total RNA input and delivers more data in less time than other technologies. Unlike conventional miRNA test methods, the FlexmiR miRNA assay does not require sample manipulation such as enrichment or amplification. And, with liquid bead arrays delivered by xMAP technology, you get faster, more reproducible results than competing planar arrays, which are limited by solid-phase kinetics.

In a fast and straight forward procedure, the FlexmiR technology allows researchers to

  •  Biotinylate the 3’ ends of total RNA, 
  •  Hybridize the labeled RNA to Tm-normalized LNA capture probes that have been  coupled to fluorescently dyed xMAP beads,
  • Tag the captured biotinylated miRNAs with streptavidin-phycoerythrin (SAPE)
  • Analyze the samples on a Luminex 200.
