xMAP® Salmonella Serotyping Assay (xMAP SSA)

The Luminex xMAP Salmonella Serotyping Assay (SSA) is a multiplex, nucleic acid-based assay for research or epidemiological use in identifying the serotype of a Salmonella isolate.

Description

The xMAP Salmonella Serotyping Assay is a CDC3&4 developed, fast, accurate and comprehensive molecular Salmonella serotyping assay.  xMAP SSA consists of three separate tests that determine O and H antigens simultaneously, including the ability to identify some serotype-specific markers in the Additional Targets or AT test.

  • Tests for 7-O Groups, 35-H antigens & 3 additional targets
  • Built on proven Luminex technology: MicroPlex® Microspheres and Luminex 100/200™
  • Kit contains: Probe coupled beads, primers, buffer, and SAPE reporter for up to 100 samples

Workflow

How the xMAP Salmonella Serotyping Assay works:

SSA Workflow 1
Step 1: Extraction  
DNA is extracted and quantified from a Salmonella isolate using a simple 15 minute extraction method.  
SSA Workflow 2
Step 2: Multiplex Amplification 
Multiplexed PCR with biotinylated primers is performed for each of three tests included in the assay.  
SSA Workflow 3
Step 3: Bead Hybridization and Detection  
Labeled amplicon is hybridized to the appropriate oligonucleotide probe-coupled microsphere mixture and labeled with SAPE reporter.  
SSA Workflow 4
Step 4: Data Acquisition and Analysis  
The assay plate is analyzed on a Luminex 100/200™ analyzer.  
Data generated may be further analyzed via the optional Samonella Analysis Tool to determine serotype based on the Kauffman-White-Le Minor scheme.
